Clinical Toxicology Regulatory Report Drafting
Draft CTD Module 2.3 safety narratives from toxicology data and adverse events
What You Can Do
This skill organizes complex toxicology datasets, clinical adverse events, and mechanistic findings into structured, FDA/EMA-compliant safety narratives. You input raw safety data—adverse event summaries, laboratory results, animal study findings, vital signs—and receive draft safety summaries, detailed safety assessments, and expedited safety reports ready for regulatory submission. It handles IND applications, annual safety updates, clinical development safety overviews, and urgent safety communications with proper MedDRA coding and regulatory formatting.
Features
Consolidates adverse event data from parallel clinical trials into unified safety profiles with trend analysis
Transforms preclinical toxicology + limited human PK/PD into compliant 15-50 page safety sections
Year-over-year adverse event summaries with causality assessment and signal detection
Links receptor binding, metabolite data, and tissue distribution to clinical AE patterns
Rapid drafting of urgent safety communications for serious emerging signals
Structures vital signs, laboratory trends, and histopathology findings per CTD guidelines
Phase transition safety overviews (Phase 1→2, 2→3) with population-level risk stratification
Organizes clinical findings with mechanistic rationale for independent assessment committees
Example Output
Example 1: IND Safety Module Summary
Input: Phase 1 PK data (n=24), Phase 1b efficacy signals (n=12), preclinical GLP tox studies (28-day rat, 90-day dog), emerging QTc signal in 3 subjects.
Output:
Safety Summary – Section 2.3.5
Nonclinical Safety: 28-day rat study (GLP) at doses up to 100 mg/kg revealed no organ toxicity. 90-day dog study showed reversible hepatocyte hypertrophy at ≥50 mg/kg, not observed at 10 mg/kg. No genotoxicity in AMES or micronucleus.
Clinical Safety: 36 subjects across Phase 1/1b. Most common AE: headache (n=8, 22%), nausea (n=6, 17%), Grade 1-2. One Grade 2 QTc prolongation (475 ms) in subject receiving 400 mg; resolved upon dose reduction. No deaths, SAEs, or withdrawals due to safety.
Example 2: Annual Safety Update
Input: 12-month ongoing Phase 2b trial data (n=187), cumulative AE database, MedDRA coding, causality assessments.
Output:
Annual Safety Update – Year 1
AE Incidence: Overall AE rate 68% (127/187), consistent with year 1 baseline (69%). No new safety signals detected. GI AEs (n=34, 18%) remain most common, unchanged from pre-specified monitoring threshold. Serious AEs: 3 hospitalizations (pneumonia, appendicitis—unrelated; 1 Grade 3 drug-related rash).
Trend Analysis: QTc signal from IND phase resolved; mean QTc change from baseline –2 ms (95% CI: –8 to +4 ms).
